(Boston) A study conducted at Boston University School of Medicine (BUSM) demonstrates an effective combination therapy for breast cancer cells in vitro. The findings, published in the July 2012 issue of Anticancer Research, raise the possibility of using this type of combination therapy for different forms of breast cancer, including those that develop resistance to chemotherapy and other treatments.

The study was led by researchers at the Boston University Cancer Center. Sibaji Sarkar, PhD, adjunct instructor of medicine at BUSM, is the study's corresponding author.

According to the Centers for Disease Control and Prevention, breast cancer is the most common cancer among women in the United States aside from non-melanoma skin cancer. Breast cancer also is one of the leading causes of cancer death among women of all races and Hispanic origin populations.

Triple negative breast cancer, which accounts for approximately 14 to 20 percent of all breast cancer cases, is a type of the disease that occurs when the cancer cells lack hormone receptors, including the receptor called HER-2, and typically will not respond to hormone and herceptin-based therapies. Triple negative breast cancer occurs more often in African-American women and is considered to be a more aggressive form of the disease with higher rates of recurrence and mortality than other forms of breast cancer.

"Cancer is like a car without brakes. Cell growth speeds up and it doesn't stop," said Sarkar. "When expressed, tumor suppressor genes, which work in a protective way to limit tumor growth, function as the brakes. They are not expressed in most cancers, causing the cancer to grow and potentially metastasize."

A major focus in the area of anti-cancer drug development is to find a way to re-express tumor suppressor genes so that they can help inhibit cancer cell growth. Some tumor suppressor genes are imprinted, meaning that from the two genes inherited from the mother and father, only one of the genes is functional. In cancer, both imprinted tumor suppressor genes may become non-functional and unable to stop tumor growth.

The researchers tested, in vitro, a combination therapy of an epigenetic drug with a protease inhibitor on breast cancer cell lines that are hormone responsive and breast cancer lines, like triple negative, that are not hormone responsive. They utilized histone deacetylases inhibitors (HDACi) and calpeptin, which inhibits calpain, a protein involved in the regulation of signaling proteins. Calpain inhibition is being studied as a potential treatment model for blood clots and other neurological diseases.

In this study, they found that the combination therapy both inhibited cell growth and increased cell death in both cancer cell lines by inducing cell cycle arrest and cell death. However, the mechanism of how the combination therapy stops the cells from growing was different. Cells in the hormone responsive line stopped the cell cycle in an earlier phase compared to the non-hormone responsive cells. In the triple negative breast cancer cell line, the inhibitors allowed an imprinted tumor suppressing gene, ARHI, to re-express, which helped stop the growth of the cancer cells and led to cancer cell death.

"The study data demonstrates that HDACi's bring back the brakes of the car, halting cell growth and promoting cell death," added Sarkar, who also is a faculty member at the Genome Science Institute at Boston University. "These results provide a model to investigate the re-expression of tumor suppressor genes, including imprinted genes, in many forms of breast cancer."

This study needs further investigation but raises the possibility of using this type of combination therapy for diverse types of breast cancers including those that are hormone refractory and develop drug resistance to conventional chemotherapy.

Whitney Houston Tribute at BET Awards (Videos)

The BET Awards show tribute to the amazing Whitney Houston is just the latest in a long list of amazing tributes to honor a fabulous woman and singer. It was quite a night in Los Angeles last night with the fabulous BET Awards. Although there were a number of outstanding performances what clearly made the evening was the tribute to Whitney Houston. Seriously the collaboration between Cissy Houston, Mariah Carey, Monica and Brandy just to name a few was spectacular. There was not a dry eye in the house when Whitney?s mom Cissy belted out an amazing version of ?Bridge Over Troubled Water? in honor of her daughter. Mariah Carey kicked off the tribute talking to the audience about her friendship with Houston and just how much she missed her dear friend. I was bawling at that point I have to be honest with you. Then you had Monica singing I Love The Lord,” Brandy belting out two? of Whitney?s biggest hits “I Wanna Dance With Somebody” and “I’m Your Baby Tonight.? There was also Chaka Khan lighting up the stage with “I’m Every Woman.” Whitney?s brother Gary Houston also performance and she was honored by her ?Waiting To Exhale? [...]

Canon confirms PowerShot S100 lens error, offers free repair for affected cams

Select PowerShot S100 cameras are encountering a lens error, Canon confirmed on its Product Advisories page. An undisclosed number of the high-end point-and-shoots have had issues with a "disconnected part inside the camera," which causes the optic to malfunction. Canon suggests that the issue may be connected with exposure to heat or humidity, but regardless of the cause, the company is offering free repairs, even for out-of-warranty cameras -- assuming the issue is caused by this specific part. Qualifying S100s should have a serial number that begins with any number ranging from 29 though 41 (29xxxxxxxxxx, for example), and this specific offer only covers residents of the US and Puerto Rico, though owners in other countries should be able to reach out to their local support centers for assistance. You'll find the full advisory at the source link below.

Poetry Anaylsis: 'The Second Coming' by William Butler Yeats ...

By Arthur Christopher Schaper Created: July 1, 2012 Last Updated: July 2, 2012

?

The Second Coming
Turning and turning in the widening gyre
The falcon cannot hear the falconer;
Things fall apart; the centre cannot hold;
Mere anarchy is loosed upon the world,
The blood-dimmed tide is loosed, and everywhere
The ceremony of innocence is drowned;
The best lack all conviction, while the worst
Are full of passionate intensity.

Surely some revelation is at hand;
Surely the Second Coming is at hand.
The Second Coming! Hardly are those words out
When a vast image out of Spiritus Mundi
Troubles my sight: a waste of desert sand;
A shape with lion body and the head of a man,
A gaze blank and pitiless as the sun,
Is moving its slow thighs, while all about it
Wind shadows of the indignant desert birds.

The darkness drops again but now I know
That twenty centuries of stony sleep
Were vexed to nightmare by a rocking cradle,
And what rough beast, its hour come round at last,
Slouches towards Bethlehem to be born
? William Butler Yeats (1865-1939)

Yeats?s poem, written in 1919, exemplifies the dilemma of the modern world, one in which man has been allowed to discard the principles, the myths, the legends, and the culture which defined his experience and informed his identity in a world he did not create.

Now pressed to create himself, to define his path in a fallen world which still cries out that something is coming, he is left with no knowledge, no wisdom, no legacy, and no authority to explain to him what is happening.

A broad, sometimes comic, yet fully harrowing aspersion of what is to come, Yeats?s dire prediction of a future where ?the centre cannot hold,? where authority and order give way to decay and chaos, contends that the rumblings of modern moral relativism have dire consequences.

At the outset, Yeats presents an unstable and foreboding phenomenon, a ?gyre,? or a circular or spiral form; a vortex, magnified by the dizzying repetition of ?turning and turning.?

?The falcon cannot hear the falconer.? Yeats incorporates medieval imagery in the second line, a trice to the glancing eye, yet inevitably stored with implications. The traditions, the myths of the past, for example, no longer withstand the onslaught of the widening vortex which disrupts and destroys everything in its wake. ?Falcon? incorporated in ?falconer? speaks to the essential identity and relationship of leader and led, yet their connection no longer remains intact.

?Things fall apart??so banal a turn of phrase in contrast to sweeping ocean currents and the romance of the hunt, yet arresting for the fundamental scope of decay which wreaks havoc on order. ?The centre? could refer to commerce, to the focal point of a ministry, the direction of a ship?a ?centre? which accounts for the integrity of the whole, which is not.

Almost a sly joke, ?mere anarchy? implies that the disorder of the modern age is weak in spite of its damage intensity on the world around it. The agent of this anarchy is not identified, perhaps weakening its power, yet also enhancing the forbidding menace of its force.

The ?ceremony of innocence? is a rich phrase, one encapsulating the traditions of men which create a sense of security in a world fraught with doubts and growing questions. Yet in a perverse twist, the ceremony itself is drowned, echoing the religious practice of baptism, losing its value, the essence never resurging.

?The best lack all conviction.? ?Conviction? speaks not just of a confident sense of the truth, but also of the internal mandate that they must do something. They feel no pang about letting the world slip into crude emptiness.

?The worst are full of passionate intensity.? The play on words with ?passionate? evokes a deep irreverence, perhaps conjuring the image of Christ?s ?Passion,? which he endured with intense conviction, whereas now, according to the poet, the ?worst,? the evil elements of the world are now committed, but their commitment is to a heedless, hedonist hatred of all order, a moral relativism which scorns at dying for anything.

The inarticulate nature of the vision illustrates the maddening disturbance of the poet?s fears. Some ?Spiritus Mundi,? a world-spirit, Weltgeist or Zeitgeist.

The poet then glimpses a mirage: ?A shape with lion body and the head of a man? conjures up the tombs of ancient Pharaohs. Yet this ?shape,? indefinite in its mixed description, ?is moving,? with birds flocking around it. Raw power, vain attempts to shade one?s authority, the awesome size and scope of ancient artifacts, have moved many to read the past in order to justify their present circumstances, to enshrine their life and worldview on something which smacks of permanence.

?Darkness drops again,? alludes, perhaps, to the modern age, with its mixing of metaphors, the inability to explain the trials and troubles of a world which has discarded the distinctions of right and wrong, or authority and submission.

Without question, Yeats cannot end his dire anti-dirge but with a question, a wandering wondering, which pieces together a narrative from times and deeds immemorial, yet lost through decay and disrespect.

GOP: Voters will have final say on health care law

(AP) ? Republican congressional leaders said Sunday that voters ? not the Supreme Court ? will have the final word on President Barack Obama's health care law come November. And they are betting that the law's unpopularity will be enough to drive Democrats from power.

The White House's response? Bring it on.

"We've got one last chance here to beat Obamacare, and we can do that in the November election," said Senate Majority Leader Mitch McConnell, calling the law the "single worst piece of legislation" passed in modern times.

White House chief of staff Jack Lew countered that he believes most Americans want to put the health care debate to rest.

"I actually think the American people want us to focus on the economy, on creating jobs and moving forward," said Lew.

Republicans and Democrats have been wrangling for the upper hand in the health care debate since last week's Supreme Court ruling upholding the law's mandate that individuals buy health insurance or face a penalty. Chief Justice John Roberts, a conservative, provided the pivotal vote in that decision by ruling that the penalty was legal under the government's taxing authority.

While technically handing a political victory to Obama, Roberts' ruling invigorated Republicans eager to cast the law as a new tax.

"The American people do not want to go down this path," said House Majority Leader John Boehner, R-Ohio. "They do not want the government telling them what kind of insurance policy they have to buy, and how much they have to pay for it, and if you don't like it we're going to tax you."

Democrats refute the characterization of the law, even as Roberts handed them a technical victory by voting to uphold the law. Lew said the mandate would impact only 1 percent of Americans ? those who can afford health insurance but refuse to buy it.

"It's a penalty for free riders," said Sen. Chuck Schumer, D-N.Y.

Yet public opposition to the health care law remains high. Forty-seven percent of respondents in a recent Associated Press-GfK poll said they oppose the law while 33 percent said they support it.

However, much of the polling does find strong support for individual elements, like allowing young adults to remain on their parents' plan to age 26. Some Democrats see that as an opening to reframe the debate.

Republicans say they believe last week's ruling by the high court gives them fresh ammunition to attack Obama ? and the Democrats who support him ? in the upcoming election because of the health care bill's connection to jobs. The GOP says the law puts onerous mandates on industry that could stifle job creation.

"I think it's intertwined with the economy, and I think it's an example where Washington doesn't get it," said Sen. Tom Coburn, R-Okla., of the health care bill.

House Minority Leader Nancy Pelosi said Sunday that Republicans, including Mitt Romney, are "being the mouthpiece of the health care industry" and that the bill will actually improve the economy.

"The costs were unsustainable," she said of health care. "It's a competitiveness issue for business and for our economy. So we had to (come) to a place where we lowered costs to all concerned, and that we again take it down a path where we continue to lower costs."

If given control of the Senate next year, McConnell said he would support using budget reconciliation rules to repeal the health care law. Doing so would prohibit Senate filibusters and require only 51 votes to succeed. In 2010, Republicans lambasted Democrats for relying on these rules to pass the health care bill, calling their tactics unusual and hyperpartisan.

McConnell said he'll do whatever it takes to repeal the law.

"I'm confident they're going to give us the votes to repeal it," he said of the American public.

The House is scheduled to vote to overturn the law on July 9. The vote will largely be symbolic since the Democrats control the Senate. But it will put lawmakers on record for the upcoming political campaign.

McConnell and Lew spoke on "Fox News Sunday." Boehner, Schumer and Coburn spoke on CBS' "Face the Nation." Pelosi spoke on NBC's "Meet the Press."
